St. Lawrence of Rome (225–258 AD) was one of the seven deacons of the Roman Church, serving under Pope Sixtus II with responsibility for the Church's material goods and the distribution of alms to the poor. Born in Huesca, in the Roman province of Hispania Tarraconensis, he was appointed Archdeacon of Rome.

During the persecution of Christians by Emperor Valerian in August 258, Pope Sixtus II was executed on 6 August. Four days later, on 10 August 258, Lawrence was also martyred. When the Roman prefect demanded that Lawrence surrender the Church's treasures, he asked for three days, distributed as much Church property to the poor as possible, then presented the poor, the widowed, and the orphaned to the prefect — declaring them to be the true treasures of the Church.

Lawrence was sentenced to death by being roasted alive on a gridiron. According to tradition, he told his executioners: "Turn me over; I am done on this side." This composure made him patron saint of cooks, chefs, comedians, deacons, the poor, and firefighters. His feast day, 10 August, is also the date chosen for the formal opening of St. Lawrence's Church in Karachi. The Perseid meteor shower, which peaks around this date, is known in folk tradition as "the tears of St. Lawrence."

The history of St. Lawrence's Church is inseparable from the history of Cincinnatus Town, one of Karachi's earliest planned townships. The neighbourhood was named after Cincinnatus Fabian D'Abreo (24 September 1862 – 25 January 1929), a prominent Goan Catholic civic leader whose father, Manuel D'Abreo, had migrated to Sindh from Goa in 1846.

The town was designed by Pedro D'Souza, a civil engineer educated at St. Xavier's College in Bombay, and his colleague George Britto. D'Souza designed the colony with St. Lawrence's Church at its centre — regarded as one of the best-planned districts of Karachi. The city later honoured the planners by naming Pedro D'Souza Road and Britto Road after them.

During the British colonial period, Goans in Karachi held positions throughout the judiciary, port authority, police, railways, postal services, and healthcare. The Karachi Goan Association, whose sandstone building dates to 1886, had approximately 1,300 members in the 1940s. Following Pakistan's independence in 1947, and particularly after the Islamization policies of the 1970s, many Goan Catholic families gradually emigrated to the United States, Canada, Great Britain, and Australia.

The parish was officially established in 1912. It was the planners of Cincinnatus Town who first gave priority to the building of a church, which they dedicated to St. Lawrence. Although the groundbreaking took place in 1903, the construction was an entirely community-funded effort that unfolded over more than two decades.

A formal building fund was started in 1907. By a contributors' meeting held in 1925, Rs. 14,500 had been collected. The church was designed by Fr. Heras, S.J. (a Jesuit priest), incorporating elements of Mughal architecture — a distinctive choice that gave the church its unique character among Catholic churches in South Asia.

Construction was completed in 1929 and the first High Mass was celebrated that same year — remarkably, before the building had a roof or doors. Parishioners brought their own stools and chairs. In 1930, parishioner Lawrence Cajetan Duarte donated a life-size statue of the patron saint, specially ordered from Rome. The church was formally opened and consecrated on 10 August 1931, the feast day of St. Lawrence.

The Catholic mission in Karachi was managed by the Society of Jesus (Jesuits) until 1934, when it was transferred to the Order of Friars Minor (Franciscans), who arrived from the Netherlands on 13 January 1934. The first parish priest was Fr. Priscus Nieuwenhuis, O.F.M. (1931–1937).

During the Second World War, the Portiuncula Friary grew vegetables for its own use and supplied St. Patrick's and St. Lawrence's parishes with fresh produce. The Franciscans even operated a cabin in the St. Lawrence's compound, patronised by parishioners after Mass. When the Archdiocese of Karachi was erected on 20 May 1948 and elevated on 15 July 1950, diocesan priests took over management from the Franciscans.

The church celebrated its Golden Jubilee in 1981, a year made especially momentous when Pope John Paul II visited Karachi and offered Mass before approximately 100,000 faithful at the National Cricket Stadium. A plaque commemorating the jubilee of the church was blessed by the Pope and placed in the walls of the church.

The 1980s represented a watershed period for the parish, marking the beginning of the final wave of emigration among the original settler families — predominantly of Goan descent — who departed for Canada, the United States, and Australia. The palatial houses of Cincinnatus Town were gradually demolished to make way for apartment buildings.

The parish celebrated its Platinum Jubilee in 2006, marking 75 years since the church's formal opening. This milestone was documented in the book Gold to Platinum (1981–2006) by Menin Rodrigues, published in 2016 as a sequel to Prof. Oswin Mascarenhas's earlier work, The Origin and Evolution of St Lawrence's Parish, which covered the period from the early 1900s to 1981.

St. Lawrence's Church is architecturally distinctive among Catholic churches in Pakistan and South Asia for its incorporation of Mughal architectural elements. Rather than the conventional Gothic or Romanesque forms typical of Western church design, the building features a prominent dome and a minaret, each topped with a cross — a striking synthesis of Islamic architectural vocabulary and Christian symbolism.

The sanctuary is positioned directly beneath the dome, creating a sense of vertical grandeur. Two galleries run along the sides of the church. High above each side of the sanctuary are three stained glass windows, with the Star of David depicted in the centre window — a design element with deep Judeo-Christian symbolism. The life-size statue of St. Lawrence, donated by Lawrence Cajetan Duarte and ordered from Rome in 1930, remains a prominent feature of the interior.

The church has been officially heritage-listed by the Sindh government as a building of historical and architectural significance.

Born in Bombay to a Goan family, Cordeiro moved to Karachi in 1926 and served as an altar boy at St. Lawrence's Church while attending St. Patrick's High School. Ordained in 1946, he was appointed Archbishop of Karachi in 1958.

In 1973, Pope Paul VI elevated him as Pakistan's first and only cardinal. He attended both papal conclaves of 1978 and was briefly considered as a potential candidate for the papacy by Time magazine after the death of Pope John Paul I. He served as President of the Catholic Bishops' Conference of Pakistan from 1958 until his death in 1994. The parish has in total produced one cardinal, two archbishops, and three bishops.
